CMS makes your job easier as web developer. There are ready to use themes and templates. Also gives you a central place to manage content of your page easily. There are more to say. However negative side is you are bound to stuff that are already written for you. Although you have control for what you need but you never going to have 100% control of your site.
Drupal, Joomla, Wordpress and plone all are CMS. Depends what you want to use it for. For example WordPress is made for making blogs, although recent versions are full fledged CMS and you can use to build almost any kind of site.
Best thing to do is to decide what kind of site you are going to make and have a research on CMS and decide which one suits you best.
The positive side of open source is that everyone has access to source code which makes more programmers contribute toward project, also programmers share their thought and make applications more secure and robust.
MySQL is an open source relational database system management.
MySQL has its own limitations depending on which engine you are using, but it does the job.
I personally use MSSQL and I feel limitted when I have to work with MySQL. For example MyISAM engine does not enforce referential integrity and I hate it. (This is absolutely personal and many MySQL users will not agree with me)
I don't see any negative side for open source programs.
